一、总RNA的定义
总RNA=mRNA+tRNA+rRNA，即：总RNA就是指mRNA、tRNA、rRNA的总和。这个概念是在还没有发现microRNA的时候的产生的，却被各大RNA提取试剂盒生产公司所默认，一直延续至今。也就是说在市场上你看到的“总RNA提取试剂盒”，只对mRNA、tRNA、rRNA负责，提出来的是这三种RNA的混合物，重点是对mRNA负责。所以各位可以去各大公司的网站，看一看总RNA提取试剂盒案例所配的RNA电泳图图，只有代表“总RNA”的2条带——28s和18s；代表microRNA的5s带，要不没有，要不很弱。这就是目前各个公司总RNA提取试剂盒中“总RNA”的概念。

那么Trizol呢？trizol也是总RNA提取试剂呀？ Trizol提出来的可是3条带吧？这该是真正的总RNA了吧？想知道答案，请接着往下看…

Based on our results, we proposed that some miRNAs are selectively destabilized depending on the adhesion status of the cells. However, in subsequent studies, we discovered that structured miRNAs with low GC content are selectively lost during sample preparation, particularly when a small number of cells is used for RNA preparation using the standard TRIzol protocol (see the Letter to the Editor published in this issue for details). These findings provide an alternative explanation for our original data. While the original data are all reproducible, we are retracting the paper because we feel the main conclusions have been compromised. We apologize for any inconvenience this may have caused.

所以， Trizol不能作为microRNA提取的首选试剂，也不能作为全转录组测序该选用的真正的总RNA提取试剂。Trizol只能适用于公司概念中的“总RNA”。

三、全转录组测序该选什么样的RNA提取试剂盒呢？
全转录组测序主要是测mRNA和microRNA。
Trizol会丢失很多microRNA，所以不能用。
总RNA提取试剂盒的重点是mRNA，根本不考虑microRNA的回收率，连RNA胶图都出不来5s带，更不能考虑啦。
